T lymphocytes play a crucial role in the pathogenesis of rheumatoid arthritis(RA). In
this study we have analyzed the sequence of complementarily-determining region
3(CDR3) of T cell receptor(TCR) V¥â17 gene expressed in the CD4+ T cell population
of synovial fluid(SF) infiltrating T lymphocytes from RA patient Unstimulated CD4+ T
cells were isolated from peripheral blood mononuclear cells(PBMC) and synovial fluid
mononuclear cells(SFMC) using maganetic beads coated with a monoclonal antibody. We
obtained TCR V¥â17 sane by the amplification of cDNA from CD4+ T cells derived
from PBMC and SFMC using reverse transcription-polymerase chain reaction(RT-PCR).
The analyses of TCR V¥â17 CDR3 in the CD4+ T cell has revealed that the amino
acids sequences of CDR3 have conserved motif in SFMC, but not in PBMC. The
analysis of CDR3 showed specific amino acid motif within N-D-N region of TCR V¥â17
derived from SFMC. Glycine was frequently used in D¥â regions, and serine and
threonine which have hydroxyl group were noticeable among the SFMC clones. These
amino acids might directly contact a specific antigen peptide for the protection of
synovitis in RA patients.
