백서의 실험적 간 섬유증식에서 면역억제 및 증강제에 의한 비장세포 Subset 변화에 관한 연구

Abstract
요 약
간 섬유화의 발생에 비장의 역할이 중요하다는 보고들이 있다. 그러나 그 기전에 관하여는
아직 불명한 점이 많다. 저자는 비장이 간 섬유화에 관여하는 기전의 일단을 구명할 목적으
로 이 연구를 시도하였다.
정상 흰쥐에 돼지혈청을 복강내에 주사하여 간 섬유화을 일으키는 실험에 있어서 돼지혈청
단독, 이것과 Freund's adjuvant, 또는 cyclosporin A를 함께 투여하여 간 섬유화의 정도 및
비장세포 subset의 비율변동을 비교 검토하였다. 이렇게 하여 얻은 성적을 요약하면 다음과
같다.
1) 간 섬유화는 돼지혈청 단독 투여에 있어서 주입시작 후 6주째와 8주째에 각각 5마리 중
3마리에서, 10주째에 5마리 중 4마리에서, 그리고 12주째에 5마리 전부에서 경하게 또는 현
저하게 나타났고, 돼지혈청과 adjuvant 투여군에 있어서 4주째에 5마리 중 1마리에서 그리
고 6주째에서 12주째까지에 5마리 전부에서 경하게 또는 현저하게 나타났으며, 돼지혈청과
cyclosporine 투여군에 있어서 8주째에 5마리 중 2마리에서 고리고 10주째와 12주째에 5마
리 중 1마리에서 경하게 나타났다.
2) 비장의 조직학적 소견에 있어서 돼지혈청 단독투여군에서는 주입시작 후 6주째에 중심
동맥 주위백수가 확대되었고 배중심이 출현하여 8추 이후에는 점차 조금씩 심하여졌고, 돼
지혈청과 adjuvant 투여 군에서는 4주째부터 비슷한 소견이 나타나 점차 심하여졌으며, 돼
지혈청과 cyclosporine 투여군에서는 별 변동이 인정되지 아니하였다.
3) 비장세포 subset 비율의 변동에 있어서 B-세포 비율은 돼지혈청 단독 투여군에서는 주
입시작 후 8 주째부터 감소하였고, 돼지혈청과 adjuvant 투여군에서는 별 변동이 없었으며,
돼지혈청과 cyclosporine 투여군에서는 4주째부터 현저하게 감소하였다.
CD4+ 세포 비율은 돼지혈청 단독 투여군과 돼지혈청과 cyclosporine 투여군에서는 주입후
4주째와 6주째에 감소하였으나, 돼지혈청과 adjuvant 투여군에서는 별 변동이 없었다. CD8+
세포 비율은 돼지혈청 단독 투여군과 돼지혈청과 cyclosporine 투여군에서는 8주 이후부터
증가하였으나, 돼지혈청과 adjuvant 투여군에서는 별 변동이 없었다. 비장 대식세포 비율은
돼지혈청 단독 투여군과 돼지혈청과 cyclosporine 투여군에서는 별 변동이 없었으나, 돼지혈
청과 adjuvant 투여군에서는 주입시작 후 초기(4주째와 6주째)에 심하게 감소하였다.
이상의 성적에 의거하면 돼지혈청에 의한 간 섬유화는 adjuvant에 의하여 촉진되고
cyclosporine에 의하여 억제되며, 돼지혈청에 의한 비장세포 subset 비율의 변동에 있어서
B-세포와 CD4+ 세포 비율은 감소하고 CD8+ 세포 비율은 증가하는데 이런 변동은 adjuvan
떼 의하여 촉진되고 cyclosporine에 의하여 억제되며, 대식세포 비율은 유의적 변동이 생기
지 아니하나 adjuvant에 의하여 현저하게 감소한다고 생각되었다.

Although there have been many reports about the importance of the spleen's role in
hepatic fibrogenesis, the exact mechanism is still uncertain. The author designed this
study to evaluate splenic function on hepatic fibrogenesis.
The degree of hepatic fibrosis and the population of splenocyte subsets were studied in
the experimental animal model with fibrosis produced by injecting normal swine serum
intraperitoneally into Sprague-Dawley rats. The animals were divided into three groups;
group A was subjected to injection of swine serum only, group B swine serum and
complete Freund's adjuvant, and group C swine serum and cyclosporin A. The
experimental hepatic fibrogenesis by swine serum was augumented by coinjection with
the adjuvant and inhibited by cyclosporin A.
The study of the splenocyte subset revealed increased percentages of splenic B cell and
CD4+ cell and a decreased percentage of CD8+ cell, and these changes of splenocyte
subset were also augumented by the adjuvant and inhibited by cyclosporin A. The
percent of monocytes was not significantly altered, although a tendancy of early
decrease by the adjuvant was noted.
