Abstract

¸ñ Àû: º» ¿¬±¸¸¦ ÅëÇØ TGF-¥â1¿¡ ÀÇÇØ À¯µµµÈ Àΰ£Àڱ󻸷ÀÇ Å»¶ô¸·È­ °úÁ¤¿¡¼­ ERK¿Í PPAR¥ãÀÇ ¿ªÇÒÀ» ±Ô¸íÇÏ°íÀÚ ÇÏ¿´´Ù.

¿¬±¸¹æ¹ý: Àڱ󻸷 ±âÁú¼¼Æ÷´Â DMEM/F12 (10% FBS, 1 nM E2 and 100 nM P4) Á¶°Ç¿¡¼­ ¹è¾çÇÏ¿´´Ù. ¿¬±¸ ¸ñÀû¿¡ µû¶ó TGF-¥â1 (5 ng/ml), Rosiglitazone (50 nM)¿Í PD98059 (20 ¥ìM)¸¦ ¹è¾ç¾×¿¡ ÷°¡ÇÏ¿´´Ù. Trypan-Blue¿Í hematocytometer¸¦ ÀÌ¿ëÇÏ¿© Çö¹Ì°æÇÏ¿¡¼­ ¼¼Æ÷ÀÇ °³¼ö¸¦ ÃøÁ¤ÇÏ¿´´Ù. Enzyme-linked immunosorbent assay (ELISA)¿Í western blotting ¹æ¹ýÀ» »ç¿ëÇÏ¿© ´Ü¹éÁúÀÇ ¹ßÇö Á¤µµ¸¦ °üÂûÇÏ¿´´Ù.

°á°ú ¹× °á·Ð: ¹è¾ç¾×¿¡ TGF-¥â1À» ÷°¡ÇÏ¿© ¼¼Æ÷ÀÇ Áõ½Ä Á¤µµ¸¦ ÃøÁ¤ÇÑ °á°ú TGF-¥â1ÀÌ ¼¼Æ÷ÀÇ Áõ½ÄÀ» ¾ïÁ¦ÇÏ´Â °ÍÀ» ¾Ë ¼ö ÀÖ¾ú´Ù. ¶ÇÇÑ ¹è¾çµÈ ¼¼Æ÷·ÎºÎÅÍ PGE2 ¹× prolactinÀÇ ¹ßÇöÀ» À¯µµÇÏ´Â °ÍÀ» ¾Ë ¼ö ÀÖ¾ú´Ù. ÀÌ·¯ÇÑ TGF-¥â1ÀÇ ÀÛ¿ëÀº Smad ¹× ERKÀÇ È°¼ºÈ­¸¦ ÅëÇÏ¿© ÀϾÀ» ¾Ë ¼ö ÀÖ¾ú´Ù. PPAR¥ãÀÇ ±âÁúÀÎ rosiglitazoneÀ» ¹è¾ç¾×¿¡ ÷°¡ÇÑ °á°ú TGF-¥â1¿¡ ÀÇÇÑ ¼¼Æ÷ Áõ½ÄÀÇ ¾ïÁ¦°¡ ¿ªÀüµÇ´Â °ÍÀ» ¾Ë ¼ö ÀÖ¾ú´Ù. »Ó¸¸ ¾Æ´Ï¶ó, ¼¼Æ÷ ³» ERKÀÇ È°¼º ¿ª½Ã ¾ïÁ¦ ½ÃÄ×À¸¸ç ÀÌ °á°ú PGE2¿Í prolactinÀÇ ¹ßÇöÀÌ ¾ïÁ¦ µÇ´Â °ÍÀ» °üÂûÇÒ ¼ö ÀÖ¾ú´Ù. µû¶ó¼­ º» ¿¬±¸¸¦ ÅëÇØ TGF-¥â1¿¡ ÀÇÇÑ Àڱ󻸷 ±âÁú¼¼Æ÷ÀÇ Å»¶ô¸·È­´Â Smad¿Í ERKÀÇ È°¼ºÈ­¸¦ ÅëÇÏ¿© ÀÌ·ç¾îÁö¸ç ÀÌ·¯ÇÑ °úÁ¤Àº PPAR¥ã¿¡ ÀÇÇØ ¾ïÁ¦µÊÀ» ¾Ë ¼ö ÀÖ¾ú´Ù.

Objective: To investigate the role of ERK and PPAR¥ã on the TGF-¥â1 induced human endometrial stromal cell decidualization in vitro.
Method: Endometrial stromal cells are cultured under the following condition: DMEM/F12 (10% FBS, 1 nM E2 and 100 nM P4). TGF-¥â1 (5 ng/ml), Rosiglitazone (50 nM), and PD98059 (20 ¥ìM) were added according to experimental purposes. Trypan-Blue and hematocytometer were utilized to count cell number. Enzyme-linked immunosorbent assay (ELISA) and western blotting were utilized to detect proteins.

Result: TGF-¥â1 inhibited proliferation of cultured human endometrial stromal cells and induced expression of PGE2 and prolactin. This effect was mediated by Smad and ERK activation. Administration of rosiglitazone, PPAR¥ã agonist,prevented TGF-¥â1 effect on cell proliferation. Furthermore, Rosiglitazone inhibited TGF-¥â1 induced activation of ERK, consequently reduced PGE2 and prolactin production.

Conclusion: TGF-¥â1 induced decidualization of endometrial stromal cell through Smad and ERK phosphorylation.
PPAR¥ã acts as a negative regulator of human ndometrial cell decidualization in vitro.
