Abstract

¸ñ Àû: º» ¿¬±¸¸¦ ÅëÇØ GnRH ÀÇÇÑ ¼¼Æ÷ ºÐ¿­ÀÇ ¾ïÁ¦´Â integrin, FAK ¹× ERK¸¦ ÅëÇÑ ¼¼Æ÷³» ½ÅÈ£Àü´Þ ±âÀüÀ» ÅëÇÏ¿© ÀϾÀ» ±Ô¸íÇÏ°íÀÚ ÇÏ¿´´Ù.

¿¬±¸¹æ¹ý: ¿¬±¸¿¡ »ç¿ëµÈ Àΰ£Àڱ󻸷¾Ï ¼¼Æ÷ÁÖ´Â DMEM/F12 (10% FBS)ÀÇ Á¶°Ç¿¡¼­ ¹è¾ç ÇÏ¿´´Ù. GnRH-I°ú -II´Â ½ÇÇè ¸ñÀû¿¡ µû¶ó 100 nM ³óµµ·Î 0, 5, 10, 15, 20, 30ºÐ°£ ¶Ç´Â 10 nM or 100 nMÀÇ ³óµµ·Î 20ºÐ°£ ó¸® ÇÏ¿´´Ù. ¼¼Æ÷ÀÇ ºÐ¿­ Á¤µµ´Â [3H] thymidine incorporation assay¸¦ ÀÌ¿ëÇÏ¿© Á¤·®ÀûÀ¸·Î ÃøÁ¤ ÇÏ¿´À¸¸ç, Immunoblotting ¹æ¹ýÀ» ÀÌ¿ëÇÏ¿© ´Ü¹éÁúÀÇ ¹ßÇöÀ» È®ÀÎ ÇÏ¿´´Ù.

°á °ú: GnRH-I°ú -II ¸ðµÎ HEC1A ¼¼Æ÷ÀÇ ¼¼º¸ºÐ¿­À» ¾ïÁ¦ÇÏ¿´À¸¸ç integrin ¥â3ÀÇ ¹ßÇöÀ» Áõ°¡ ½ÃÄ×´Ù. GnRH-I°ú -II¸¦ ó¸® ÈÄ FAK ¹× ERKÀÇ ¾È»êÈ­°¡ Áõ°¡µÊÀ» °üÂûÇÒ ¼ö ÀÖ¾ú´Ù.

°á ·Ð: GnRH¿¡ ÀÇÇÑ ¼¼Æ÷ºÐ¿­ÀÇ ¾ïÁ¦´Â integrinÀÇ ¹ßÇö°ú FAK ¹× ERKÀÇ ÀλêÈ­ °úÁ¤À» ÅëÇÏ¿© ÀϾÀ» ¾Ë ¼ö ÀÖ¾ú´Ù.

Objective: To investigate new signal transduction cascade through integrin, FAK and ERK in the suppressed cell proliferation by GnRH-I and -II.

Method: Human endometrial cancer cells (HEC1A) were cultured under the following condition: DMEM/F12 (10% FBS). GnRH-I and -II were treated time (0, 5, 10, 15, 20, 30 min; 100 nM) and dose (10 nM or 100 nM; 20 min) dependent manner according to experimental purposes. Cell proliferation was measured using [3H] thymidine incorporation assay. Immunoblotting was utilized to detect proteins.

Results: GnRH-I and -II inhibited proliferation of HEC1A cells and induced expression of integrin ¥â3. Phosphorylation of FAK and ERK were induced by GnRH-I and -II.

Conclusion: GnRH inhibited cell proliferation via the expression of integrin and FAK, ERK phosphorylation.
