Purpose : Recently, the cellular biology of response to hyperthermia was understood
and localizing hyperthermic equipments were developed. And the results of hyperthermic
treatment were reported in several malignant tumors, but the effect of hyperthermia on
advanced renal cell carcinoma(RCC) is poorly understood. We investigated the effect of
hyperthermia on growth and DNA synthesis of RCC cell line in vitro.
Materials and Methods : Human renal cell carcinoma cell line, Caki-1, was used.
Hyperthermia(41.5¡£C and 43¡£C) was applied with various duration(1, 2, and 4 hours).
The cell growth was measured by MTT colorimetric assay. The 24-hour DNA
synthesis was studied with 3H-thymidine uptake test. The effect of
hyperthermia on cell-cycle was analysed by flowcytometry.
Results : In 2-hour treatment with 41.5¡£C or 43¡£C of hyperthermia, the cell growth
was inhibited up to 25% and in 4-hour treatment up to 50% The DNA synthesis was
inhibited as similar pattern of cell growth. In flowcytometry, the decrease of S-phase
fraction in hyperthermia treated cells was recognized in dose and time dependent
manner.
Conclusions : These results suggest that the localizing hyperthermia could be applied
In advanced renal cell carcinoma patients for cytoreductive purpose because there are
yet no definitive treatment modalities.
