Abstract

¸ñÀû: ÀڱñÙÁ¾ÀÇ Å©±â º¯È­¿¡´Â ¼¼Æ÷ºÐ¿­¿¡ ÀÇÇÑ ¼ºÀåÀÌ¿Ü¿¡µµ ¼¼Æ÷ÀÇ °£ÁúÀÇ ±¸¼º ¼ººÐÀÇ º¯È­¿Í remodeling¿¡ ÀÇÇÑ ¼ºÀå±âÀüÀÌ °ü¿©ÇÑ´Ù. ÀÌ·¯ÇÑ ¼¼Æ÷ÀÇ °£ÁúÀÇ ±¸¼º¼ººÐÀÇ »ý¼º°ú ÃàÀû¿¡ °ü¿©ÇÏ´Â matrix metalloproteinase(MMPs)¿Í tissue inhibitor of metalloproteinase(TIMPs)ÀÇ ¹ßÇöµµ¸¦ »ìÆ캸°í ¼¼Æ÷ÀÇ °£ÁúÀÇ ´ë»ç¿¡ °ü¿©ÇÏ´Â ÀÎÀÚµéÀ» ±Ô¸íÇÏ¿© ÀڱñÙÁ¾ÀÇ ¼ºÀå¿¡ ¹ÌÄ¡´Â ¿µÇâÀ» ¾Ë¾Æº¸°íÀÚ ÇÏ¿´´Ù.
¿¬±¸¹æ¹ý: ÀڱñÙÁ¾À¸·Î Áø´ÜµÇ¾î ÀÚ±ÃÀûÃâ¼úÀ» ½ÃÇàÇÏ¿© 30¿¹ÀÇ Á¤»ó Àڱà ±ÙÃæ Á¶Á÷°ú Àڱà ±ÙÁ¾ Á¶Á÷À» ¾ò¾î RNA¸¦ ÃßÃâÇÏ¿© MMPs´Â collagenase, 72kDa gelatinase, 92kDa gelatinase, matrilysin, stromelysin-2,-3, mRNA¸¦, tissue inhibitors´Â TIMP-1, -2, -3 mRNA¸¦ °ËÃâÇϱâ À§ÇÑ primer¸¦ Á¦ÀÛÇÏ¿© ¿ªÀü»ç ÁßÇÕÈ¿¼Ò ¹ÝÀÀ(Reverse transcriptionpolymerase chain reaction, RT-PCR)À» ½ÃÇàÇÏ¿© ±× ¹ßÇöµµ¸¦ ºñ±³ ºÐ¼®ÇÏ¿´´Ù.
°á°ú: Àڱà ±ÙÁ¾°ú Àڱñ٠¸ðµÎ¿¡¼­ collagenase, stromelysin, gelatinaseÀÇ mRNA°¡ ´Ù ¹ßÇöµÇ¾ú´Ù. 92kDa gelatinase mRNAÀÇ ¹ßÇöµµ°¡ °¡Àå ³·¾Æ ¹Ì¹ÌÇÏ°Ô ¹ßÇöµÇ¾ú°í, stromelysin-3 mRNAÀÇ ¹ßÇöµµ´Â ÃÑ 30¿¹Áß 25¿¹¿¡¼­ ÀڱñÙÁ¾¿¡¼­ Àڱñٿ¡ ºñÇØ ³ôÀº ¹ßÇöµµ¸¦ º¸¿´À¸¸ç(P<0.05), ´Ù¸¥ MMPs mRNAÀÇ ¹ßÇöµµ´Â Àڱà ±ÙÁ¾°ú Á¤»ó Àڱñٰ£¿¡ Â÷ÀÌ°¡ °ÅÀÇ ¾ø¾ú´Ù. ÀÌ·¯ÇÑ stromelysin-3 mRNAÀÇ ¹ßÇöµµÀÇ Â÷ÀÌ´Â ºÐºñ±â¿¡ ´õ ÇöÀúÇÑ Â÷À̸¦ º¸¿´À¸¸ç(P<0.05), Àڱà ±ÙÁ¾ÀÇ ¹«°Ô°¡ »ó°ü°ü°è°¡ ¾ø¾ú´Ù. ¶ÇÇÑ Àڱà ±ÙÁ¾°ú Àڱñ٠¸ðµÎ¿¡¼­ TIMP-1, -2, -3 mRNA°¡ ¹ßÇöµÇ¾ú´Ù. ±×Áß TIMP-3 mRNAÀÇ ¹ßÇöµµ°¡ ÃÑ 30¿¹Áß 26¿¹¿¡¼­ Àڱà ±ÙÁ¾¿¡¼­ Àڱñٿ¡ ºñÇØ ³·Àº ¹ßÇöµµ¸¦ º¸¿´À¸¸ç(P<0.05), ÀÌ·¯ÇÑ Â÷ÀÌ´Â »ý¸®Áֱ⳪ Àڱà ±ÙÁ¾ÀÇ ¹«°Ô°¡ »ó°ü°ü°è°¡ ¾ø¾ú´Ù.
°á·Ð: Àڱà ±ÙÁ¾°ú Á¤»ó ÀڱñÙÀÇ ¼¼Æ÷¿Ü °£ÁúÀÇ ±ÕÇü¿¡´Â metalloproteinase¿Í tissue inhibitors°¡ °ü¿©ÇÏ´Â °ÍÀ» ¾Ë ¼ö ÀÖ¾úÀ¸¸ç, metalloproteinaseº¸´Ù´Â tissue inhibitors°¡ ´õ ÁÖ¿äÇÑ ÀÛ¿ëÀ» ÇÏ¸ç ±× Áß¿¡¼­µµ TIMP-3 mRNA°¡ Á¾¾ç¾ïÁ¦ÀÎÀÚ·Î ´õ Áß¿äÇÑ ÀÛ¿ëÀ» Çϸç Àڱà ±ÙÁ¾À¸·Î Á¾¾çÈ­ µÇ¸é¼­ stromelysin-3°¡ ¼¼Æ÷¿Ü °£ÁúÀÇ ÃàÀûÀ» °¡Á®¿Í ¢¥fibrotic tumor¢¥ÀÇ ¼ºÁúÀ» °¡Á®¿À°Ô ÇÏ¸ç ºÐºñ±âÀÇ ÁÖÈ£¸£¸óÀÎ ÇÁ·ÎÁ¦½ºÅ×·ÐÀÇ ¿µÇâÀ» ¹Þ¾Æ Àڱà ±ÙÁ¾ÀÇ ¼ºÀåÀ» °¡Á®¿À´Â °ÍÀ¸·Î »ç·áµÈ´Ù.
Objectives: To determine the level of mRNA expression of various members of the matrix metalloproteinase and tissue inhibitors in uterine leiomyoma compared with unaffected myometrium.
Materials & Method: 30 cases of portions of leiomyoma and myometrium were collected immediately following hysterectomy. Thirteen cases were from proliferative phase and seventeen were from secretory phase of menstrual cycle. The mean age was 43.7years old. The level of expression of mRNAs of interstitial collagenase, gelatinise, stromelysin, TM¢¥-1, 2,-3 was determined by reverse transcriptase-polymerase chain reaction(RT PCR) and normalized to GAPDH(glyceraldehyde-3-phosphate dehydrogenase) mRNA.
Results: Myometrium and leiomyoma expressed all the members of above mentioned matrix metalloproteinase family and tissue inhibitors. Leiomyoma expressed a significantly higher level of stromelysin-3 during secretory phase, an extremely lower level of 92kDa gelatinase and a significantly lower level of TAMP-3. The immunohistochemical localization of TAMP-3 was smooth muscle cell and arteriole wall of myometrium and leiomyoma.
Conclusions: The increased expression of stromelysin-3 in uterine leiomyoma compared with myometrium suggests that this MMP may be involved in the formation of a more fibrous extracellular matrix in leiomyoma. The extremely lower expression of 92kDa gelatinase of leiomyoma means that leiomyoma do not invade myometrium and forms a separated mass. Decreased expression of TM-3 of leiomyoma suggests that TM-3 is required for differentiation and homeostasis of extracellular matrix of normal myometrium and function as a suppressive role of tumor development
