Abstract

¸ñÀû : ¿¬·É°ü·ÃȲ¹Ýº¯¼º¿¡ À־ ¿©·¯ ¹ß»ý ÀÎÀÚ Áß ¸Á¸·»ö¼Ò»óÇǼ¼Æ÷ÀÇ »ç¸êÀÌ °ü·ÃµÈ °ÍÀ¸·Î ¾Ë·ÁÁ® ÀÖ´Ù. º» ¿¬±¸¿¡¼­´Â Àڿܼ±ÀÌ ¸Á¸·»ö¼Ò»óÇǼ¼Æ÷ÀÇ »ç¸êÀ» À¯¹ßÇÏ´ÂÁö ¾Ë¾Æº¸°í À̶§ ¹ÝÀÀ»ê¼Ò±â°¡ Áõ°¡µÇ´ÂÁö ¾Ë¾Æº¸°íÀÚ ÇÏ¿´´Ù.
´ë»ó°ú ¹æ¹ý : ¹è¾ç¸Á¸·»ö¼Ò»óÇǼ¼Æ÷¸¦ well-plate¿¡ ÀÏÁ¤ÇÑ ¼ö·Î ³Ö°í 24½Ã°£ µ¿¾È ºÎÂø½ÃŲ ÈÄ Àڿܼ±À» 0-80 mJ/§²ÀÇ ¾çÀ¸·Î Á¶»çÇÏ¿´´Ù. Á¶»ç ÈÄ ÀÏÁ¤ÇÑ ½Ã°£ °£°ÝÀ¸·Î »ýÁ¸ ¼¼Æ÷¼ö¸¦ °æÁ¤ÇÏ°í annexin V, propidium icdide(Pt)¸¦ ÀÌ¿ëÇÑ À¯¼¼Æ÷ºÐ¼®À» ½ÃÇàÇÏ¿© apoptosisÀÇ Á¤µµ¸¦ °áÁ¤ÇÏ¿´´Ù. ¶ÇÇÑ ¼¼Æ÷³» ¹ÝÀÀ»ê¼Ò±âÀÇ ¹ß»ý·®À» DCFHDA¸¦ ÀÌ¿ëÇÏ¿© Á¤·®È­ÇÏ¿´´Ù.
°á°ú : Àڿܼ±ÀÇ Á¶»ç·®ÀÌ Áõ°¡ÇÒ¼ö·Ï ¸Á¸·»ö¼Ò»óÇǼ¼Æ÷ÀÇ »ç¸êÀÌ Áõ°¡ÇÏ¿© 40 mJ/§²ÀÇ ¾çÀ¸·Î Á¶»çÇÑ °æ¿ì¿¡ 24½Ã°£ÈÄ 20~30%ÀÇ ¼¼Æ÷ »ç¸êÀÌ ÀϾ´Ù. À¯¼¼Æ÷ºÐ¼®¿¡¼­ annexin V ¾ç¼º ¼¼Æ÷ÀÇ ºÐÀ²ÀÌ Á¶»ç ÈÄ ½Ã°£ÀÌ °æ°úÇÒ¼ö·Ï Áõ°¡ÇÏ¿´´Ù. ¶ÇÇÑ Àڿܼ±À» Á¶»çÇÑ °æ¿ì¿¡ ¹ÝÀÀ »ê¼Ò±â°¡ Áõ°¡ÇÏ¿´´Ù.
°á·Ð : ÀÌ»óÀÇ °á°ú´Â Àڿܼ± Á¶»ç¿¡ ÀÇÇÑ »êÈ­ ¼Õ»óÀÌ ¸Á¸·»ö¼Ò»óÇǼ¼Æ÷ÀÇ »ç¸ê ¿øÀÎÀ¸·Î Áß¿äÇÑ ¿ªÇÒÀ» ÇÒ °¡´É¼ºÀ» ½Ã»çÇÑ´Ù.

Purpose: The death of retinal pigment epithelial(RPE) cells plays an important role in the development of age-related macular degeneration. We studied whether ultraviolet could induce apoptotic cell death and the increase of reactive oxygen intermediates (ROI) in the RPE cells.
Methods: Cultured human RPE cells were exposed to doses of between 0 and 80 mJ/cm2 UVB as an attached monolayer. The number of viable cells was assessed by trypan blue assay. The apoptosis was determined by flow cytometry using annexin V and propidium iodide (PI). Intracellular ROI was measured using DCFH-DA.
Results: UVB-exposed cells showed a dose-dependent cell death. The number of cells decreased in 20?30% at the dose of 40 mJ/cm2 UVB 24 hours after UVB exposure. The proportion of annexin V positive cells and intracellular ROI increased after UVB exposure.
Conclusions: These results suggest that UV induced oxidative damage may be one of the important mechanisms of RPE cell death.
