Abstract

¸ñÀû : º£Å¸ÀÛ¿ëÁ¦°¡ ¼¶À¯ÁÖ¼¼Æ÷ÀÇ »ýÁ¸°ú NOÀÇ »ý¼º¿¡ ¾î¶² ¿µÇâÀ» ¹ÌÄ¡´ÂÁö ¾Ë¾Æº¸°íÀÚ ÇÏ¿´´Ù.
´ë»ó°ú ¹æ¹ý : µÅÁöÀÇ ¼¶À¯ÁÖ¼¼Æ÷¸¦ ÀÏÂ÷¹è¾çÇÑ ÈÄ º£Å¸Â÷´ÜÁ¦ÀÎ timolol°ú propranololÀ» °¢°¢ 10, 50, 100, 500 §­°ú 0.1, 1, 10§­ÀÇ ³óµµ·Î, ±×¸®°í º£Å¸ÀÚ±ØÁ¦ÀÎ isoproterenolÀ» 0.1, 1, 10 §­ÀÇ ³óµµ¿¡ 1ÀÏ, 3ÀϾ¿ °¢°¢ ¶Ç´Â µ¿½Ã¿¡ ³ëÃâ½ÃÄ×´Ù. ¼¼Æ÷ÀÇ »ýÁ¸À²Àº MTT assay¸¦ ÀÌ¿ëÇÏ¿´°í, Griess assay¸¦ ÀÌ¿ëÇÏ¿© ¹èÁöÀÇ nitrite ¾çÀ» ÃøÁ¤ÇÏ¿´´Ù.
°á°ú : °¢ ¾à¹°ÀÇ ³ëÃâÀº ¼¼Æ÷ÀÇ »ýÁ¸¿¡´Â À¯ÀÇÇÑ ¿µÇâÀ» ³¢Ä¡Áö ¾Ê¾Ò´Ù. TimololÀº 50 §­ºÎÅÍ NOÀÇ »ý¼ºÀ» À¯ÀÇÇÏ°Ô ¾ïÁ¦ÇÏ¿´À¸¸ç(p<0.05), propranololµµ NOÀÇ »ý¼ºÀ» ¾ïÁ¦ÇÏ¿´´Ù. IsoproterenolÀº timololÀÇ È¿°ú¸¦ À¯ÀÇÇÏ°Ô Â÷´ÜÇÏ¿´´Ù.(p<0.05). ÀÌ·¯ÇÑ °á°ú´Â 1ÀÏ ±×¸®°í 3ÀÏ°£ÀÇ ³ëÃâ¿¡¼­ À¯»çÇÑ °á°ú¸¦ ³ªÅ¸³»¾ú´Ù.
°á·Ð : º£Å¸Â÷´ÜÁ¦ÀÎ timolol°ú propranololÀº ¼¶À¯ÁÖ¼¼Æ÷¿¡¼­ NOÀÇ »ý¼ºÀ» ¾ïÁ¦ÇÏ¿´À¸¸ç ÀÌ´Â isoproterenol¿¡ ÀÇÇØ »ó¼âµÇ¾ú´Ù. µû¶ó¼­ º£Å¸ÀÚ±ØÁ¦´Â ¼¶À¯ÁÖ¼¼Æ÷¿¡¼­ NO»ý¼º¿¡ °ü¿©ÇÔÀ» ¾Ë ¼ö ÀÖ¾ú´Ù.

Purpose: To investigate the effect of ¥â-adrenergics on the production of nitric oxide (NO) in cultured trabecular meshwork cells.
Methods: Primarily cultured porcine trabecular meshwork cells were exposed to timolol and to propranolol at various concentrations. Cellular survival was assessed by MIT assay and the production of nitrite was assessed by Griess reaction after 24 hours and 3 days respectively. Also investigated was the effect of co-administraton of timolol and isoproterenol.
Results: After exposure for 24 hours, neither drug affected the cellular survival. Timolol and propranolol inhibited the production of NO significantly (p<0.05). Isoproterenol abolished timolol-induced inhibition of NO production after 24 hours. These results were similar after exposure for 3 days.
Conclusions: Both timolol and propranolol inhibit the production of NO in trabecular meshwork cells and isoproterenol abolished this effect. These results suggest that ¥â-adrenerics involves in the production of NO in trabecular meshwork cells..
