Abstract


A wide-spectrum initiation model was investigated in female N: GP(S) mice. Sequential treatments with benzo(a)pyrene (BP), dimethylnitrosamine(DMN), with or without N-methylnitrosoures(MNU), resulted in tumor formation in the lungs in 84.2%~90.0%
of
animals, but did not produce any tumorous lesions in other organs. The lung tumors were mainly adenomas and 4 cases showed carcinoma in adenoma. The mean number of adenoma was 13.2~16.2 per mouse. MNU combination had no additive effect on the
lung
tumor
incidence, but significantly increased the lung tumor multiplicity. The carcinogen-treated animals showed cytomegaly of hepatocytes and this was more marked in MNU combined group. These results showed that a wide-spectrum initiation model was not
established in this mouse system. Further study with a change in mouse strain or in combination of carcinogen, or increment of carcinogen dose would be necessary.
