INTRODUCTION
Somatic gene therapy is a rapidly developing therapeutic approach for the treatment of
human diseases, especially in cancers. Use of suicide gene is one way to treat cancers.
The gene product of the herpes simplex virus thymidine kinase (HSVtk) gene, the most
widely used suicide gene, converts nontoxic nucleotide analog, ganciclovir (GCV), into a
highly logic phosphorylated GCV that acts as a chain terminator of DNA synthesis and
an inhibitor of DNA polymerase, selectively killing dividing cells. Such a strategy has
been used to treat experimental brain tumors through in vivo gene transfer with
retroviral-vector-producer cells. Subsequent in vivo applications of HSVtk gene with
GCV injection resulted in tumor reduction in several animal models although retroviral
HSVtk transduction of every cell is impossible. To explain the death of non-transduced
tumor cells, a bystander effect was hypothesized, where HSVtk expressed from cells is
able to cause the death of neighboring col]s that do not themselves express the gene.
Several mechanisms were proposed to explain the bystander effect; phagocytosis of
material from dying HSVtk expressing cells and gap-junctional intercellular
communication.
In this study, we compared two hepatocellular carcinoma cell lines in vitro with
different growth characteristics, one anchorage-dependent and the other
anchorage-independent cell line, by expressing HSVtk gene cloned onto a retroviral
vector and by measuring their sensitivity to GCV and the bystander effect.
