In inflammation of the respiratory mucosa, the adhesion molecules play important roles
in cell-to-cell interaction, especially cellular migration and infiltration. Intercellular which
expresses at the luminal surface of the epithelium and endothelium is a major ligand for
leukocyte function-associated antigen(LFA)-1, ¥â2 integrin on the
leukocyte, therefore expression of ICAM-1 on the epithelium has been reported to be
potential importance in the pathogenesis of allergic respiratory diseases such as
bronchial asthma and allergic rhinitis. Because of difficulty in cell culture of the nasal
epithelium, the ICAM-1 expression on the nasal epithelium has not been investigated
clearly. The aim of this study Is to investigate the kinetics of ICAM-1 expression on
the cultured human nasal epithelial cells(HNEC) by cytokines.
The authors cultured the HNEC by monolayer culture of dissociated cell method and
analyzed ICAM-1 expression after incubation with cytokines using flow cytometry
(Fluorescence Activated Cell Sorter, FACS) with anti-ICAM-1 monoclonal antibody.
The following results were obtained. The cultured HNEC expressed ICAM-1
constitutively. ICAM-1 expression was unregulated by tumor necrosis factor-¥á (TNF-
¥á), interferon-¥ã (IFN-¥ã) but not by in terleukin-l¥á (IL-l¥á). IFN-¥ã was more
effective for the unregulation of ICAM-1 than TNF-¥á in a dose-dependent manner and
had synergistic combination with TNF-¥á. The ICAM-1 expression was unregulated at
6 hours alter incubation with IFN-¥ã (1000/m1) and the maximum expression was
observed at 24 hours.
The authors hope that the culture method and these results could be used as basic
data for further study to investigate the role of the epithelial cell, ICAM-1 and
cytokines in the inflammatory nasal mucosal diseases.
