Abstract
요약
목적 : 강력한 단백분해효소 억제제인 gabexate mesilate가 기저 상태와 cholecystokinin
자극 상태에서 췌장 외분비에 미치는 영향을 파악하고자 본 연구를 시행하였다.
대상 및 방법 : 백서를 12시간 금식 후 마취하에 개복하여 담췌관을 PE-10으로 분리후
내경정맥을 통하여 생리식염수 (1.56 mL/hr), gabexate mesilate (30 mg/kg) 및 CCK-8
(3.9 pmol/hr)를 투여한 후 췌장액을 채취하여 췌장액 분비량, 중탄산염량 및 단백량을 측정
하였다.
결과 : 췌장액 분비량은 기저군에서 20.1±1.1 μL/30 min, gabexate mesilate 투여군에서
17.1±1.6 μL/30 min으로 기저군과 비교시 유의한 차이가 없었다. CCK-8 투여군에서는
58.9±3.9 μL/30 min, CCK-8 자극 후 gabexate mesilate 투여군 56.2±4.0 μL/30 min,
gabexate mesilate 투여 후 CCK-8 자극군에서는 54.3±3.8 μL/30 min로서 세 군간에 유의
한 차이가 없었다. 췌장액 중 중탄산염량은 기저군에서 30.1±1.3 rmol /30 min, gabexate
mesilate 투여군 29.6±1.6 μmol/30 min으로 차이가 없었으며, CCK-8 투여군에서는 193.1
±4.3 μmol/30 min, CCK-8 자극 후 gabexate mesilate 투여군 185.±6.7 μmo1/30 min,
gabexate mesilate 투여 후 CCK-8 자극군 183.2±5.6 μmo1/30 min로서 중탄산염량 또한
세 군간의 유의한 차이는 없었다. 단백량은 기저군에서 39.8±1.2 μg/30 min이었으나
gabexate mesilate 투여군에서는 30.1 ±1.3 μg/30 min으로 유의한 감소가 있었으며, CCK
투여군에서는 396.0±6.2 μg/30 min, CCK-8 자극 후 gabexate mesilate 투여군 295.9±5.9
μg/30 min, gabexate mesilate 투여 후 CCK-8 자극군 310.5±7.4 μg/30 min로서 후자 양
군간에서는 유의한 차이는 없었으나, CCK-8 투여군과 비교시 유의한 감소가 있었다.
결론 : Gabexate mesilate가 백서의 췌장 외분비능에 미치는 영향으로는 췌장액 분비량과
중탄산염량에는 관련이 없고 기저 상태 및 CCK 자극 상태에서 단백량을 감소시키는 것을
확인할 수 있었다.

Background/Aims : Gabexate mesilate, a serine protease inhibitor, is a synthetic
compound which is not antigenic and diffuses easily We have investigated the effect of
gabexate mesilate on exocrine pancreatic secretion in rat at basal and
cholecytokinin-stimulated state.
Methods : After anesthetized male Sprague-Dwaley rats were incised in the midline of
abdomen, PE-10 tube was inserted into the pancreatic duct. The secretion was collected
after the ligation of the bile duct between pancreas and liver The volume, of pancreatic
secretion and the amount of bicarbonate and protein in pancreatic secretion were
measured in basal and CCK-8 (3.9 μmol/hr) stimulated state with and without the
administration of gabexate mesilate (30 mg/kg).
Results : The volume was 20.1±1.1 μL/30 min in basal state and 58.9±3.9 μL/3O
min in CCK-8 stimulated state. The amount of bicarbonate in basal and CCK-8
stimulated state was 30.1±1.3 μmol/ 30 min and 193.1±4.3 μmo1/30 min, respectively.
After infusion of gabexate, the volume and the amount of bicarbonate were not changed
significantly. The amount of protein was 39.8±1.2 μg/30 min in basal state and 396.0±
6.2 μg/30 min in CCK-8 stimulated state. After the infusion of gabexate mesilate, the
amount of the protein in basal and CCK-8 stimulated state was significantly decreased
to 30.1±1.3 μg/30 min and 295.9±5.9 μg/30min, respectively (p<0.05).
Conclusions : With the infusion of gabexate mesilate in both basal and CCK
stimulated state of rat, only the protein in pancreatic exocrine secretions was
significantly decreased.
