-Abstract-
Background/Aims: Adenoma and adenocarcinoma of the ampulla of Vater are frequently
associated. We investigated to see whether p73 gene could play a role as a potential
tumor suppressor gene in this adenoma-carcinoma sequence in the ampulla of Vater.
Methods: Tissues of 31 ampulla of Vater cancers, 24 adenomas, 10 metastatic lymph
nodes, and 35 normal regions were collected and microdissected under a microscope.
DNA was extracted from the tissues and mutations in exon of p73 gene were detected
by polymerase chain reaction-single strand conformation polymorphism and direct
sequencing.
Results: Four base substitutions were detected in p73 gene analysis. One mutation was
observed in the intron (C to T) region of one metastatic lymph node. Two mutations
observed in adenocarcinoma portion of two patients showed no amino acid substitution.
mutation showed amino acid substitution (Alanine to Threonine) at codon 156 in
adenoma.
Conclusions: The mutations of p73 gene are rare in both adenoma and carcinoma of the
an ampulla of Vater It is unclear whether p73 gene plays a role as a tumor suppressor
gene.
