Abstract

¸ñ Àû: Àú»ê¼ÒÁõÀº ¹æ»ç¼± °¨¼ö¼ºÀ» ÇöÀúÈ÷ °¨¼Ò½ÃÅ°¸ç, ÀÌ¿¡ ´ëÇÑ ÀûÀÀ ¹ÝÀÀ¿¡¼­ hypoxia-inducible factor 1¥á (HIF-1¥á)°¡ Áß¿äÇÑ ¿ªÇÒÀ» ÇÏ°í ÀÖ´Ù. HIF-1¥áÀÇ ¹ßÇö°ú ¹æ»ç¼± °¨¼ö¼º°úÀÇ »ó°ü °ü°è¸¦ ¾Ë¾Æº¸°íÀÚ ÇÏ¿´´Ù.

´ë»ó ¹× ¹æ¹ý: ÁãÀÇ °£¾Ï ¼¼Æ÷ÁÖÀÎ hepa1c1c7 ¼¼Æ÷¿Í HIF-1¥â°¡ °á¼ÕµÇ¾î HIF-1¥áÀÇ ±â´ÉÀÌ ¾ïÁ¦µÈ hepa1C4 ¼¼Æ÷¸¦ »ç¿ëÇß´Ù. °á °ú: Hepa1c1c7 ¼¼Æ÷¿¡¼­´Â DFX Àüóġ¸¦ ÇÑ °æ¿ì ¹æ»ç¼±¿¡ ÀÇÇØ HIF-1¥áÀÇ ¹ßÇöÀÌ Áõ°¡ÇßÀ¸³ª, hepa1C4 ¼¼Æ÷ÁÖ¿¡¼­´Â º¯È­°¡ ¾ø¾ú´Ù. Hepa1C4 ¼¼Æ÷ÀÇ ¹æ»ç¼± °¨¼ö¼ºÀº DFX󸮿¡ µû¸¥ ¿µÇâÀÌ ¾ø¾úÀ¸³ª hepa1c1c7 ¼¼Æ÷ÀÇ ¹æ»ç¼± °¨¼ö¼ºÀº DFX¸¦ ÀüóġÇßÀ» ¶§ À¯ÀÇÇÏ°Ô °¨¼ÒÇÏ¿´´Ù.

°á ·Ð: Àú»ê¼Ò À¯»ç ¹°ÁúÀÎ DFX¿¡ ÀÇÇØ À¯µµµÈ HIF-1¥á°¡ ÁãÀÇ °£¾Ï ¼¼Æ÷ÁÖ¿¡¼­ apoptosis¿Í ¹æ»ç¼± °¨¼ö¼ºÀ» °¨¼Ò½ÃÄ×´Ù. ÀÌ·¯ÇÑ °á°ú´Â Á¾±«³»ÀÇ Àú»ê¼Ò ¼¼Æ÷¿¡¼­ ¹æ»ç¼±¿¡ ÀÇÇØ HIF-1¥á°¡ À¯µµµÇ°í ÀÌ·Î ÀÎÇØ Àú»ê¼Ò ¼¼Æ÷¿¡¼­ ¹æ»ç¼± °¨¼ö¼ºÀ» ÀúÇϽÃÅ°´Â °ÍÀ¸·Î »ý°¢µÇ¾ú´Ù.

Purpose: It is well known that the radiosensitivity of tumor cells can be significantly reduced under hypoxic conditions. Hypoxia-inducible factor-1¥á (HIF-1¥á) plays a pivotal role in the essential adaptive responses to hypoxia. Therefore this study investigated the relationship between HIF-1¥á expression and radiosensitivity.

Materials and Methods: Mouse hepatoma cell line hepa1c1c7 and HIF-1¥â-deficient mutant cell line hepa1C4 were used to analyze the role of HIF-1¥á on radiosensitivity. These cells were exposed for 6 h to desferrioxamine (DFX) before radiation. HIF-1¥á expression was examined by Western blot. Apoptosis was assessed by DNA fragmentation, propidium iodide staining, and apoptotic cell death detection ELISA kit. Radiation sensitivity was determined using MTT assay. The radiobiological parameters, surviving fractions at 2 Gy and 8 Gy, and mean inactivation dose (MID) from the linear-quadratic model were used to assess radiation sensitivity in the statistical analyses.

Results: The expression of HIF-1¥á was increased, whereas apoptosis was decreased, by radiation in the presence of DFX in hepa1c1c7, but not in hepa1C4. The radiosensitivity of hepa1C4 cells was not significantly affected by DFX treatment. The radiosensitivity of hepa1c1c7 cells was significantly decreased in the presence of DFX

Conclusion: The expression of HIF-1¥á by hypoxia-mimic agent DFX reduced apoptosis and radiosensitivity in mouse hepatoma cell line hepa1c1c7. These results suggested that HIF-1¥á could be induced by irradiation in hypoxic cells of tumor masses, and that this might increase radioresistance in hypoxic cells.
